Nanoemulsion formulation: A novel approach to improving the bioavailability of theophylline and its potential in the management of asthma.
A nanoemulsion formulation (NEF) of theophylline was prepared and characterized. NEF was prepared by suspending the drug in the oil phase using the ultrasonication method. The effect of formulation and process variables on globule size, viscosity and pH was studied. NEF was evaluated for drug content uniformity, in vitro drug release, chemical stability, and in vitro cytotoxicity. In vitro drug release was done using USP dissolution test apparatus II. Cytotoxicity of the NEF was performed by MTT assay on NCI-H460 lung cancer cell line. The NEF of theophylline had a globule size of 186 ± 0.31 nm, viscosity of 1.59 ± 0.01 mPa.s, and pH of 4.38 ± 0.01. The drug content was 96 ± 2.12%. In vitro drug release studies revealed that NEF has a sustained release property. At the end of 8 h of dissolution, 92% drug was released from the NEF, as compared to only 8% drug release from the physical mixture. Drug release from NEF followed Higuchi model. The cytotoxicity studies suggested that the NEF was non-cytotoxic to the NCI-H460 cells.
